Gabapentin: Side Effects, Dosage, How to use

What is Gabapentin?

Gabapentin is an anti-epileptic drug, also called anticoagulant. It affects the chemicals and nerves in the body, which are prone to seizures and some types of pain.It is used in adults to treat neuropathic pain (nerve pain) caused by herpes virus or herpes zoster (herpes zoster).

Neurontin's brand of gabapentin is also used to treat seizures in adults and children who are at least 3 years old.

The Horizant brand of gabapentin is also used to treat restless legs syndrome (RLS). Just use the brand and form of gabapentin prescribed by your doctor. Each time you refill, check your medication to make sure you get the correct form.

The active ingredient in capsule, pill and oral solution Neurontin is gabapentin, the chemical name of 1- (amino ethyl) cyclo hexaacetic acid.

The molecular formula is Gabapentin C9H17NO2 and the molecular weight is 171.24.

Gabapentin is a white to off-white crystalline solid with a pKa1 of 3.7 and a pKa2 of 10.7. It is freely soluble in water and both basic and acidic aqueous solutions.

Each Neurontin capsule contains 100 mg, 300 mg, or 400 mg of gabapentin and the following inactive ingredients: lactose, cornstarch, talc, gelatin, titanium dioxide, FD&C Blue No. 2, yellow iron oxide (300 mg and 400 mg only), and red iron oxide (400 mg only). The log of the partition coefficient (noctanol/ 0.05M phosphate buffer) at pH 7.4 is –1.25.

Each Neurontin tablet contains 600 mg or 800 mg of gabapentin and the following inactive ingredients: poloxamer 407, cornstarch, copovidone, magnesium stearate, hydroxypropyl cellulose, talc, and candelilla wax

Neurontin oral solution contains 250 mg of gabapentin per 5 mL (50 mg per mL) and the following inactive ingredients: glycerin, xylitol, purified water, and artificial cool strawberry anise flavor.

Maximum dose: 50 mg / kg per day.

Dose of children (age 0-2 years)

Drug use is not available for people under the age of 3 years.

Overdose (65 years and older)

Your kidney function may decrease with age. Your body may get rid of this drug slowly. Your doctor may start you at a lower dose, so that you do not get too much of this medicine in your body. Excessive medication in your body can be dangerous. Your doctor may change your dose based on how your kidneys work.

Symptoms of over dose

A lethal dose of gabapentin was not detected in mice and rats receiving only an oral dose of 8000 mg / kg. Symptoms of acute toxicity in animals include ataxia, respiration, ptosis, sedation, hypothyroidism, or irritation.

Gabapentin can be removed by hemodialysis. Although several prescriptions for hemodialysis have not been performed, they may be seen in the clinical situation of the patient or in patients with renal impairment.

It is reported that acute overdose of NEURONTIN is up to 49 grams. In these cases, dual vision, numbness, drowsiness, numbness and diarrhea were observed. All patients recovered with supportive care. In patients with chronic renal failure treated with NEURONTIN, coma has been reported to be resolved with dialysis.

Some side effects in children with gabapentin are more likely. If your child has one of the following side effects, contact your doctor:

Change in behavior;

Memory problems;

Focus problem; or

Restless, hostile or aggressive.

Common side effects of gabapentin may include:

Headache, dizziness, drowsiness, fatigue;

Swelling in your hands or feet;

Problems with your eyes;

Coordination problems or

(In children) fever, nausea, vomiting.

This list is not the full list of side effects and others may occur.

Drugs that may interact with Gabapentin

Other drugs may interact with gabapentin, including prescription and over-the-counter medications, vitamins and herbal products. Tell your doctor about all your current medications and any medication you are taking or not taking.

Using gabapentin with other drugs that cause of drowsiness can make this effect worse. Ask your doctor before taking a sleeping pill, opiate drugs, sedation, or medication for anxiety, depression or seizure.

Other antiepileptic drugs

Gabapentin is not significantly metabolized and is not associated with the metabolism of conventional antiviral drugs.

Reducing NEURONTIN with hydrocodone reduces hydrocodone. If you use hydrocodone in patients who start or stop NEURONTIN, the risk of exposure to hydrocodone should be taken into account.

When gabapentin is prescribed with morphine, patients should see signs of depression in the central nervous system, such as drowsiness, sedation, and respiratory depression.

Pharmaceutical / laboratory interactions

Since fetal positive reading was reported using the Ames N-Multistix SG test for urine protein testing, when gabapentin was added to other anti-epileptic drugs, a specific method for determining the amount of urine protein present has been suggested.

Maalox (Aluminum Hydroxide, Magnesium Hydroxide)

The average gain of gabapentin displacement is reduced by about 20% with the simultaneous use of Maalox containing magnesium and aluminum hydroxide. It is recommended that gabapentin be taken at least 2 hours after taking the Malvox.

Substance abuse and dependence

Controlled substance

Gabapentin is not a scheduled drug.

Gabapentin does not bind to benzodiazepine, an epidemic (hair, delta or kappa), or cantiopide 1. Few cases of gabapentin withdrawal show abusive use. These people receive excessive gobapentinic intake for unauthorized use. Most people described in these reports have a history of drug abuse or the use of gabapentin to eliminate withdrawal symptoms from other substances. When administering gabapentin, patients are carefully evaluated for the history of drug use, and for signs and symptoms of abuse or misuse of gabapentin (for example, tolerance development, accelerated drug use, and drug use behavior). Give

Rare reports are available after the sale of people with withdrawal symptoms shortly after discontinuation of the recommended dose of gabapentin used to treat diseases that are not approved. Such symptoms include disturbances, disorder and confusion after a sudden interruption of gabapentin, which was then resolved after rebooting gabapentin. Most of these people have a history of drug abuse or the use of gabapentin to eliminate withdrawal symptoms from other substances. The potential of dependence and misuse of gabapentin in human studies has not been investigated.

For whom Gabapentin might not be safe

You should not use gabapentin if you are allergic to it.

To make sure this drug is safe for you, tell your doctor if you have ever had:

Kidney disease (or if you are on dialysis);

Depression, a mood disorder, or suicidal thoughts or actions;

A seizure (unless you take gabapentin to treat seizures);

Liver disease;

Heart disease; or

(For patients with RLS) if you are a day sleeper or work a night shift.

Some people have thoughts about suicide while taking this medicine. The doctor should check your progress by regular visits. Your family or other carers should also consider changes in your mood or signs.

Use Gabapentin while pregnancy

Control of seizures during pregnancy is very important and seizure may harm the mother and the baby. Do not start before taking gabapentin without your doctor's advice, and tell your doctor quickly if you are pregnant.

Pregnancy Category C

There are no adequate and controlled studies in pregnant women. In non-clinical studies in mice, mice and rabbits, gabapentin was used when used in pregnant animals when using similar or less doses than clinical trials. NEURONTIN should be used during pregnancy only if it justifies the potential risk of a fetus.

In pregnant rats, gabapentin (500, 1000, or 3000 mg / kg / day) received oral oral doses during the period of organogenesis; embryo and embryo toxicity (an increase in the prevalence of skeletal changes) was observed at two higher doses. The dose without fetal developmental toxicity in mice was 500 mg / kg / day or about ½ the maximum recommended human dose (MRHD) was 3600 mg / kg on the body surface (mg / m²).

When pregnant rabbits were treated with gabapentin during the period of organogenesis, an increase in fetal mortality was observed at any test dose (60, 300 or 1500 mg / kg). The lowest dose effect on fetal toxicity in rabbits is less than that of MRHD.

In studies in which the oral dose of gabapentin (500-2000 mg / kg / day) in rats, during pregnancy, undesirable effects were observed on the growth of children (increased hydrant and / or hydronephrosis) at all doses. The lowest dose effect for growth toxicity in rats is approximately MRHD at mg / m².

In a published study, gabapentin (400 mg / kg / day) was injected intraperitoneal into newborn infant mice during the first week after delivery, synaptosis in rodents (related to the last trimester of pregnancy in humans). Gabapentin significantly reduced the formation of neuronal synapse in the brain of intact rats and induced abnormal neural synapses in the model of synaptic mice. Gabapentin has been proven in a laboratory that is involved with the action of a receptor involved in neuronal synaptogenesis with the activity of subunit α2δ of active-calcium canals. The clinical significance of these discoveries is unclear.

To provide information on the effects of exposure to neurotitra, doctors recommend that pregnant patients registering NEURONTIN in their pregnancy registration register.
